Just Googled it, its about 150km so depending how long you need to be there for I would hire a car, if you wanted to get buses I haven't a clue how many changes you would need or how long it would take
I knew of someone who travelled by bus from Alicante airport to Cehegin, changing at Murcia city.
The bus terminates at Caravaca I believe, but I've no idea if there is a bus from there to Moratalla or the timetables.
Also, there are trains which link Murcia city and Calasparra but the railway station is quite a way from Calasparra itself.
